What does this mean?

Word(s) \t Definition

Like this?

Word1 Word2 Word3 \t Definition

How about this?

Compound Word1 Compound Word2 Compound Word3 \t Definition

Will this work?

The two halves get dumped to the opf --- so the left-hand side follows the opf standard for words to be defined. I expect that arbitrary compounds will work, but I don't know -- it's been many years since I played with this.

Take a look at https://github.com/olety/epwing2kindle/blob/master/tab2opf.py which claims to support multiple definitions
